Firefighters from many communities are battling the blaze in central Hanover.
Hanover police say the blaze engulfed the Fabulous Fashions building and the former accommodation at the Forum Hotel.
Fire crews from Hanover, Walkerton, West Gray and Elmwood are on the scene.
Police are asking everyone to avoid the city center and much of 10th Street is closed.
